Atlas of breast cancer infiltrated B-lymphocytes revealed by paired single-cell RNA-sequencing and antigen receptor profiling

Abstract To gain mechanistic insights into the functions and developmental dynamics of tumor-infiltrated immune cells, especially B-lymphocytes, here we combine single-cell RNA-sequencing antigen receptor lineage analysis to characterize a large number triple-negative breast cancer infiltrated cells report comprehensive atlas B-lymphocytes. The transcriptional profiles reveal significant heterogeneity in B-cell subgroups. analyses demonstrate that compared with those peripheral blood, B-cells have more mature memory characteristics, higher clonality, class switching recombination somatic hypermutations. Combined suggest local differentiation within tumors. signatures based on results are significantly associated improved survival tumor patients. Functional populations mechanistically, subgroups may contribute immunosurveillance through various pathways. Further dissection will provide valuable clues for immunotherapy.
